The tube's contents should be thoroughly mixed. Close to but not touching the agar dish's outer edge, place a loop of culture broth on the agar's surface. With the loop flat against the agar, lightly scratch the inoculum; avoid digging up the agar with the loop. Allow the loop to cool after sterilisation. Make a sequence of four back-and-forth strokes on the open agar dish with your left hand, each one going through the inoculum and reaching to one side. Let the loop cool down after disinfecting it. Extend the lines from the previous set of four across and across the agar dish's surface to the opposite side, then rotate the agar dish. Afterward, flip the plate over and do the same thing again with the parallel stripes. Lastly, add some streaks to the centre of the plate. Keep the original inoculum out of reach.
