The order of procedures for Gram staining is Crystal violet-Iodine-Alcohol/acetone-Safranin.

Gram Staining

It is a procedure for differentiating between bacteria species with thick peptidoglycan cell walls and those without the feature.

The bacteria with thick cell walls are able to retain the color of the dye used in the procedure and are said to be Gram-positive while those that cannot do so are said to be Gram-negative.

The procedures for carrying out Gram staining is as follows:

  • colony of the bacterium is heat-fixed unto a clean slide
  • Crystal violet is added and rinsed off with water
  • Gram iodine is added and rinsed off with water
  • Alcohol is added and also rinsed off
  • Safranin is added, allowed to stay for about a minute, and then rinsed off
  • The slide is allowed to dry and then viewed under the microscope.
